Superoxide Generation In Catfish Liver Microsomes Treated With Menadione
Armando A. Andaya
Abstract:
The study investigated the possible involvement of oxygen free radicals in mediating the toxicity
of menadione (2-methyl-1,4-naphthoqoinone) in channel catfish lctalurus pU11cta1us L. under laboratory
exposure conditions. It was hypothesized that menadione is metabolized via a flavoenzyme
NADPH-cytochrome P-450 reductase resulting in the production of semiquinones which under aerobic
condition generar.es toxic oxygen species including superoxide anions, hydrogen peroxide, and
singlet oxygen. The probable existence of this flavoenzyme in catfish liver~ was iooica(ed
and its possible involvement in lhe metabolism of menadione was demonstral.Cd. The concentrationdependent
rates of menadione-induced cyt.ochromc c reduction and oxygen consumption provided
evidences. for lhe presence of superoxide generating system in catfish liver miaosooies. These obser•
vat.ions validare the claim that oxygen free radicals could be involved, at least in part, in the mode
of toxic action of menadione in channel catfish.
